Comparison of Highly Purified Isolation of Polymorphonuclear Granular Neutrophil by Use of Chemiluminescence


Abstract
Several different methods are currently used in numerous laboratories for the isolation of human polymorphonuclear granular neutrophil (PMN). Mothods include partial purification and hypotonic shock. It is well known that PMN are blood cells of
fundamental importance for the aoility of the animlal body to eliminate foreign particles. These are sonsitive, short-lived non-divided. Therefore, in order to study their function, it is essential to use rapid and gentle purifuication methods.
Highly purified PMN-preparations (more than 97% PMN) were isolated from the peripheral blood of healthy human donored by two-step Percoll denstiy gradient avoiding hypotonic shock treatment. Human PMN were monitored by chemiluminescence(CL0 for
their
capacity to release oxygen metabolites. Lucigenin-and luminol-enhanced CL from these PMN, as compared with Ficoll-Histopaque PMN, was more sensitive and reproducibly performed in RPMI 1640 medium containing 1% of suitable bovine serum albumin
(BSA).
